Główny wątek fabularny Director's Cut dotyczy zagadkowych morderstw, które sprawiają wrażenie, jakby były w nie zaangażowane nadprzyrodzone siły. Gracze podążają za tropem podrzuconym im przez Avę, szukając nowych poszlak na Pandorze, Promethei, Eden-6 oraz Nekrotafeyo.

Director's Cut wprowadza także tzw. Vault Cards, które otwierają graczom drogę do zdobycia pakietów tematycznych przedmiotów (znajdziemy pośród nich sprzęt będący hołdem dla poległych bohaterów z uniwersum Borderlands). Vault Cards można odkryć, aktywować, a następnie wzmocnić.
Borderlands 3: Director's Cut nie oferuje nowych trybów rozgrywki. Podstawą pozostaje kampania fabularna, która możemy przejść samotnie lub w kooperacji z trzema kolegami.
